This study investigated the potential additive effects of the SARM RAD140 on muscle hypertrophy when combined with functional overloading (FO) of the plantaris muscle in Sprague-Dawley rats. Four treatment groups were established, including RAD140-FO, RAD140-Control, Vehicle-Control, and Vehicle-FO. The rats in RAD140 groups received the drug through drinking water, while control groups received only the vehicle (methylcellulose). Muscle weight analysis revealed a significant increase in RAD140-FO groups compared to control groups, although not statistically different from the vehicle-FO group. Similar findings were observed in muscle fiber cross-sectional area (CSA). The results suggest the potential of RAD140 to stimulate muscle hypertrophy, but further investigation with longer treatment duration is needed for a better understanding of the physiological response to functional overloading and SARMs treatment.
